B5培养基是1968年由Gamborg等为培养大豆根细胞而设计的,双子叶植物在B5培养基上生长更适宜。 B5(Gamborg)培养基的主要特点是含有较低的铵,这是因为铵可能对不少培养物的生长有抑制作用。经过试验发现,有些植物的愈伤组织和悬浮培养物在MS培养基上生长得比B5培养基上要好,而另一些植物,在B5培养基上更适宜。
在2.4-D存在条件下,B5培养基用于大豆根系细胞的悬浮培养。硝酸盐的浓度在20-30mM之间, 2mM硫酸铵的额外添加能使促进细胞的生长。NH4+作为唯一的氮源时,并不能支撑植物的生长,用NH4NO3代替(NH4)2SO4时也有类似的结果。然而,当铵盐的浓度超过2mM时反而抑制生长。
培养基中磷酸盐、钙离子和镁离子浓度的变化相对地对植物生长的影响较小。硫胺素是细胞生长的必需营养物质,浓度高达10mg/l。
