  Background:

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. Histone H3 is an important protein in the emerging field of epigenetics, where its sequence variants and variable modification states are thought to play a role in the dynamic and long term regulation of genes.

Among 25 analyzed species, the sequence of the synthetic peptide used for immunization is 100% homologues with the sequence in Glycine max, Zea mays, Spinacia oleracea, Triticum aestivum, Sorghum bicolor, Physcomitrella patens, Chlamydomonas reinhardtii, Cucumis sativus, Oryza sativa, Medicago truncatula, Gossypium raimondii, Vitis vinifera, Setaria viridis, Brassica rapa, Hordeum vulgare, Nicotiana tabacum, Populus trichocarpa,and 80-99% homologues with the sequence in Brassica napus.
The sequence of the synthetic peptide used for immunization is 93% homologues with the sequence in Histone H3-like 1 (AT1G13370), Histone H3.2 (AT5G65360, AT5G10400, AT5G10390, AT3G27360, AT1G09200), Histone H3-like 2 (AT1G19890) ,and Histone H3-like 4 (AT5G12910).

Application Example

Lane 1: 3 μg nuclear protein from Arabidopsis thaliana.
Lane 2: 6 μg nuclear protein from Arabidopsis thaliana.
Electrophoresis:15% SDS-PAGE
Transfer: blotting to NC (nitrocellulose) membrane for 1h.
Blocking: 5% skim milk at RT or 4℃ for 1h.
Primary antibody: 1:1000 dilution overnight at 4℃.
Secondary antibody: 1:10000 dilution using Goat Anti-Rabbit IgG H&L (HRP) (Cat# PHY6000)
Detection: using chemiluminescence substrate and image were captured with CCD camera.
